I would look for any third-party security software you may have installed that is conflicting. In particular, if you have another software firewall, it might be conflicting with the Windows Firewall. Additionally, you might want to run a virus scan on your system.
If your system is a member of a domain, it's possible that Group Policy is turning your Windows Firewall off, in which case you need to talk to your administrator about that.
